چکیده

Numerous studies have so far been conducted on the cultivation of Mycobacterium leprae and Mycobacterium lepraemurium. However, no media-bound method of cultivation has been established. A number of investigators have reported that M. lepraemurium may be cultivated by cell culture technics. The method employed by Chang (1. 2. 3 ), Chang and Neikirk (4), Chang, Anderson and Vaituzis (~), in which mouse peritoneal macrophages were employed, may be said to have contributed much to progress in the cultivation of M. lepraemurium. In the present study M. lepraemurium enclosed in diffusion chambers implanted in" the mouse peritoneal cavity gave evidence of in vivo extracellular growth. Additional studies observed the fate of M. lepraemurium when enclosed in diffusion chambers with either mouse peritoneal macrophages or LBu cells, which are the mouse stable strain cells.

In vitro growth of Mycobacterium lepraemurium: Transition from in vivo grown to in vitro adaptation.

Mycobacterium lepraemurium (M. lepraemurium), agent of rat leprosy and an interim model of human leprosy bacillus, was first discovered by Stefansky in 1903. It was considered as an obligate intracellular parasite until 1972 when Nakamura (1972) succeeded in growing this organism in vitro in his cell-free NC-5 medium; later this was confirmed by Dhople and Hanks (1977). It has been learned that...

Growth of Mycobacterium lepraemurium in Cell-Impermeable Diffusion Chambers.

Successful growth of Mycobacterium lepraemurium has been achieved by use of a specialized diffusion chamber technique. The cell-impermeable porous chambers were maintained in animals for periods up to 50 days with and without macrophages and LM cells. A generation time of 6 to 8 days was found for the acid-fast bacilli in chambers containing macrophages when maintained in the mouse. Also, cell-...

Comparative effects of sulfones and rifampin on growth of Mycobacterium lepraemurium in macrophage diffusion chamber cultures.

A cell-impermeable diffusion chamber technique has been developed that lends itself to growth studies of Mycobacterium lepraemurium. This technique, in which the organism grows within macrophage cultures inside the chambers that are maintained on monolayer cultures of macrophages, provides a method for a strict in vitro evaluation of antileprosy drugs without the influence of a multiplicity of ...

Growth of Mycobacterium lepraemurium in Nonstimulated

Mycobacterium lepraemurium cells were found to multiply in normal mouse peritoneal-derived and bone marrow-derived macrophages in vitro. Whereas activated peritoneal-derived macrophages demonstrated marked bacteriostasis for M. lepraemurium, significant bactericidal activity was exhibited by activated bone marrow-derived macrophages. However, only a small proportion of the bacteria were killed ...

Limited in vitro multiplication of Mycobacterium leprae: application to screening potential antileprosy compounds.

Inability to cultivate Mycobacterium leprae in vitro has been regarded as the bottleneck which obstructs research on modes of transmission, pathogenesis, immunization and the treatment of leprosy . The bottleneck in the cultivation problem always has been that of gaining significant information from an organism that has given no useful response to intensive inquiries by the disciplines of bioch...
